The outer wall of the brain capillary endothelium is enclosed by pericytes and astrocyte end feet, anatomically assembled to guarantee barrier functions. We, here, focus on the pericyte modifications occurring in disease conditions, reviewing evidence supporting the interplay amongst pericytes, the endothelium, and glial cells in health and pathology.

Abstract: Central nervous system pericytes have critical and complex inductive, structural, and regulatory roles interacting with other cell types of the neurovascular unit, especially endothelial cells and astrocytes. The blood–brain barrier (BBB) constitutes a neurovascular unit formed by microvascular endothelial cells (ECs), pericytes, and perivascular astrocytes (Hawkins and Davis, 2005). The ECs of the BBB display a specialized phenotype characterized by the presence of tight junctions (TJs) that limit diffusion of water-soluble compounds from the blood to the brain tissue ( Brightman and Reese, 1969 ). Development of the blood–brain barrier (BBB) requires spatiotemporal coordination of cerebrovascular endothelial cells (ECs) and pericytes. Until now, the molecular mechanism(s) coordinating the pericyte–EC behaviors during this process have been incompletely understood.

Pericyte, Astrocyte and Basal Lamina Association with the Blood Brain Barrier (BBB) The periendothelial accessory structures of the BBB include pericytes, astrocytes, and a basal membrane. The endothelial cells of the BBB are distributed along the length of the vessel and completely encircles the lumen. Pericytes play a crucial role in the formation and functionality of the blood–brain barrier.

2020-09-09 · The blood-brain barrier (BBB), which protects the CNS from pathogens, is composed of specialized brain microvascular endothelial cells (BMECs) joined by tight junctions and ensheathed by pericytes and astrocyte endfeet. The brain pericyte–like cells induced blood-brain barrier properties in BMECs, including barrier enhancement and reduced transcytosis. Last, brain pericyte–like cells were incorporated with iPSC-derived BMECs, astrocytes, and neurons to form an isogenic human model that should prove useful for the study of the NVU. Alzheimer’s disease, blood-brain barrier, pericytes, glioma, stroke The blood–brain barrier (BBB) is a well-established and unique component of the CNS. The BBB’s unique ability to selectively permit transport of substances from the vascular supply to nervous system cells is critical to ensure brain homeostasis and prevent toxic injury to the constituent elements of the brain. Endothelial cells (ECs) that form the capillary walls constitute the physical barrier but are dependent on interactions with other cell types. In vitro models are widely used in BBB research for mechanistic studies and drug screening Pericytes surround blood vessel capillaries and endothelial cells, neurons and astrocytes within the blood-brain-barrier (BBB) neurovascular unit. Pericytes, also known as mural cells, are multipotent perivascular cells that play an important role in the development, maturation, and remodeling of blood vessels.
